N,N-Diisopropylethylamine (DIPEA): A Versatile Non-Nucleophilic Tertiary Amine

DIPEA (CAS #: 7087-68-5), aka Hünig's base, is one of the most widely used organic bases in synthetic chemistry. DIPEA's tertiary nitrogen is sterically shielded by two bulky isopropyl groups and one ethyl group, giving it strong basicity while virtually eliminating nucleophilic character. This unique combination makes it superior to other tertiary amines and an excellent acid scavenger—efficiently neutralizing HCl and other protic acids generated during coupling reactions, acylations, and alkylations without participating in unwanted side reactions. DIPEA is also compatible with a wide range of functional groups, making it well-suited for complex, multi-step syntheses. Additionally, its low water solubility simplifies product isolation through standard aqueous workup.

Applications

DIPEA is used in the synthesis of over 100 FDA-approved APIs. Key applications include:

Peptide synthesis — the base of choice in both solid-phase and solution-phase peptide coupling, activating carboxylic acids while minimizing racemization.

Acylation and sulfonylation — scavenging acid byproducts to drive reactions toward completion.

Palladium-catalyzed cross-coupling — facilitating catalyst regeneration without poisoning Pd through unwanted coordination.

Heterocyclic ring formation — enabling synthesis of pyrimidines, quinazolines, imidazoles, and benzimidazoles found in kinase inhibitors, antivirals, and CNS therapeutics.

Agrochemical synthesis — supporting sulfonylurea herbicide and other crop protection agent manufacturing.

Other applications: DIPEA has been used in the synthesis of polymers, organic semiconductors, aroma chemicals, and in other emerging areas (e.g. photocatalysis, biotech-grade RNA extraction etc). DIPEA and hydrogen fluoride (HF) complexes are powerful selective and efficient nucleophilic fluorinating reagents. DIPEA is also used as green solvents.
